No. A quote, or estimate is a non posting transaction and thus has no effect on inventory. Only changing to or starting with an invoice will put a hold on items, and then only if you have set preferences to prohibit selling items whose stock falls to zero. If you allow yourself to sell prior to having stock on hand then any hold is immaterial (unless all inventory us entirely unique never duplicated)

You can't. QBO Quote is a plain simple type, not that advanced.

Is it possible now? or at least while querying the item availability can we number of items put on hold or part of  estimates

Utilize an inventory management app to reserve stocks for sale when an Order is in the 'Active' state. This deducts stock from the total available stock. Once an order is fulfilled, the stock then leaves the 'On Hand' totals.
