It takes Enterprise "Build Assemblies" with two user logged in, 1.05 minute to build and close.
When 3 or more people log in, the time goes up to 2-3 minutes per build.
Does anyone have any ideas on how to speed up these transactions?
Switch to single user mode. Any different?
How big is your file size?
Single user is as slow, no difference.
In fact, QuickBooks suggestion was keep an extra user logged onto the server at all times.
Every Monday, at 5 am, I "Rebuild the Files", backup every morning first thing.
File Size: 440,148K

Next step, run the Condense Data utility to downsize your file. Remove your old transactions and make your file size below 200 MB. Make sure to secure a backup file. Any difference?
I have set the Condense Data utility in motion.
IT will set up a test company using the latest backup and run the Condense Data utility, if all goes well, they will run it on our company file over the next weekend.

Got the same issue, not yet checked the software. But noticed that if I navigate away from QB to say Chrome, the transactions go faster....

If you're file is a large size, I suggest using the Condense Data utility. This helps streamline your company data file.
Here are some valid reasons to Condense:
You have a large data file.
You are nearing a list limit.
You have upgraded all your hardware.
Your accountant and your technical support have discussed other options and are willing to proceed with Condensing.

Let me help share information about condensing QuickBooks company files.
QuickBooks Desktop allows you to reduce your file’s overall size and still keep all your data and details. This is with the use of the Condense feature.
In line with your question, yes you can condense data using QuickBooks Desktop Premier 2016. Know that being able to condense your file may vary on the version of QuickBooks that you're using. Older versions of QBDT tend to affect the system's performance.
Before you start condensing the company file, you can run a report to see the logic the program uses to determine which transactions it will not condense. Please take note that condensing cannot be undone at a later point. If you wish to continue, follow the steps below:

You can run the Condense Data utility on your QBD 2016 US version. But if you need to split your files, you have to run it on QBD 2019 US version or later ones.
Ok, I run QB 2016 Premier Accounts version UK....
Another trick I found out, if you click build and new, its quicker than build and close. You can still close the input screen once the build has been posted
How large should my company file be, currently its about 65 Mb.
How many items should there be on the lists? Is there a place where I can check?
My items list has about 300 items, there are more to be added over next 2 months or so.
UK 2016 so can't condense.
Have check software and OS, all seems to be fine

I'm sorry this doesn't help but it's taking me upwards of 8-10 minutes per build, which means hours dedicated to just builds. I do them in Single User Mode because if they're done while everyone else is working, they get error messages saying they can't complete their task (creating sales orders, invoices, entering bills, entering customer payments, etc.) until I finish my task (the assembly build) and since the builds take SO incredibly long, we can't afford to waste that kind of time. Sometimes, QuickBooks is the worst.

The issue which we were seeing on Win7 VDIs could be due to the Network hardware device connected with the machine. If TCP/IP scaling is not supported by the network device then the performance will be slow.
Disable auto tuning level of the TCP. Please follow below steps: 1) Open command Prompt with admin right (Run as Admin) 2) Type “netsh interface tcp set global autotuninglevel=disabled” 3) After running above command restart the machine.
