Once you set up the sales tax, QuickBooks will do tax calculations automatically. Creating a purchase order will not affect the calculation. It's because a PO is a non-posting transaction.

If you are asking on which “ Income account “ , its on “ sales “ , and for “ Expense account “ its “ Purchases “
Im not really sure if I get your point or not but however these setting is since we create an account with your system

Once you set up sales tax, QuickBooks automatically calculates the total tax rate for each sale based on the following:
An estimate/quote is a non-posting transactions which you can give to your customer for a quote, bid, or proposal for your work plan to do.
If you're using this when recording your transactions, they won't show in the Taxable Sales Summary report. The report will show the posting transactions such as invoices or when you make a sale. Thus, they will only reflect on the report after converting them into invoices.
