Since we installed the latest update to Profile (v. 2021.2.5) on January 31, 2021, we have not been able to Create a New 2021 T3 file or carry forward an existing 2020 T3 file to 2021. When we try either we get the following error:
Object reference not set to an instance of an object.
We can Create a New 2020 T3 without issue.
We are currently up to date on our licensing.
I have tested this on multiple workstations and they all have the same problem.

This is an issue that will be resolved in ProFile 2021.3.0, targeted for the week of February 14, 2022. The work around is to go to the edit tab in ProFile options, Options > Environment > Edit. In the capitalization section make Normal the selection. This should resolve the issue and allow carry forward and opening 2021 T3 returns.

My firm is having the exact same issue. I will add that I have updated Windows as well and the problem persists.

I am glad the workaround helped! The issue will be fixed in ProFile version 2021.3.0. This version will also contain the new 2021T3 e-file CRA has implemented. For more information we have an article here.
