I select YES, but the price does not update in the item database or reflected when I create a new purchase order with the same item.

As I've tried it here on my end, I was able to update the new pricing of an item when creating a purchase order. The program crashes when the version becomes incompatible with the current QuickBooks Desktop (QBDT) version. Thus, let's ensure that your QBDT version is updated to its latest release. This way, you'll always have the latest features and fixes for several problems.
Then, let's run the verify and rebuild tool. This tool will scan any damaged data and will attempt to resolve them. It allows your QuickBooks software to refresh the data inside the company file.
Also, to ensure that you have the original data you can restore at any time, I'd suggest securing a backup copy of your company file before making any changes.
Once done, here's how to verify your company data:

When you update the price of the item in the database, you'll have to make sure to click the Yes radio button on the Do you want to update the item record with the new cost and Do you want to update the price for this item based on the new cost section. This is to ensure that the updated price will reflect when you create purchased order with the same item. Otherwise, if you only tick the Yes on the cost section, it won't change the price since the two options work differently and are meant to work independently.
If the issue continues to occur, you have the option to change or edit the information on the item so the updated price appears on the transactions.

This is happening to me too in QBO. The error message I receive when I try to edit the sales price is that I am using the wrong tax code and that I should find the right one.
