I have Microsoft 365 and Quickbooks Desktop Premier 2016 on my computer, and when I try to import an Excel list of inventory items, I get an error saying Microsoft Excel could not be found. what could the issue be, compatibility?

You should be able to export the report since QBDT 2016 is compatible with Office 365. Since you've encountered an error while importing the list, it can be due to the following:
If Excel is installed on your computer and there are no issues. You can also check the format when importing list. If the issue persists, we could have tried to update QuickBooks to the latest release. However, 2016 QuickBooks is an outdated one, and it won't receive any updates.

In my Quickbooks 2016, when you create a new item list on the right side of the pop-out window there is a section where you can enter 4 customs fields and link them to be shown in the actual "quote" window. However I cannot map any custom fields in the Excel item import, just the standard fields.

I have a solution for you if you are looking to import Custom Fields into a QuickBooks Item list. We have a data import utility called Zed Axis that you can find in the Apps for QuickBooks Desktop center that does support importing Custom Field data into the Item list in batch, you can see a pre-recorded demonstration of that here as well as detailed instructions on how to do that here.
I hope that provides you a quicker way to batch import custom field data.
